Permax / Dostinex

Permax, a widely prescribed drug used to manage the various symptoms of Parkinson’s disease and Restless Leg Syndrome, was recalled in March 2007 after reports of serious heart valve damage was linked to its use. Another similar dopamine agonist, Dostinex, has also been associated with life-threatening cardiac injury. Though it’s not approved for the treatment of Parkinson’s in the U.S., Dostinex is commonly prescribed to manage certain hormonal problems (excess prolactin production). Cardiac Valvulopathy

In 2006, two studies were published in the New England Journal of Medicine finding that Permax (pergolide) and Dostinex (cabergoline) were indeed linked to cardiac valvulopathy—a condition in which the mitral, atrial, and tricuspid heart valves leak. One study concluded that nearly one-fourth of Parkinson’s patients who used Permax or Dostinex suffered some type of moderate to severe heart valve damage. The other report indicated that users of these drugs faced almost seven times the risk of experiencing leaky heart valves than those who took other Parkinson’s drugs.

 

Currently, 14 Parkinson’s patients who have taken Permax have had to undergo heart valve replacement surgery to correct the damage. As a result of these serious risks, Eli Lilly and Co., the makers of Permax, have pulled the drug from the U.S. market.
